I was wondering if anyone/everyone would be willing to offer their opinion on what make, length, or boat is the quintessential custom Carolina sportfishing yacht.

Some well-know builders would include; Ritchie Howell, Ricky Scarborough, Sculley, Paul Mann, Briggs Boatworks, Blackwell etc.

Some newer builders, Spencer, Bayliss, Weaver, Winter, OBX, Jarrett Bay, etc.

If not a boat, what would be some characteristics that are important that not all builders adhere to?

I have worked with several of the builders you name. In the early 90's a 42-45 foot boat was the average. They are getting bigger and 65-75 is common.
